In this article

ASCO 2026 Preview: Key Studies to Be Announced in Chicago from 29 May - 2 June

The American Society of Clinical Oncology (ASCO) Annual Meeting is the world’s largest oncology congress; it takes place at McCormick Place in Chicago each year in late May and early June, with more than 40.000 delegates. The 2026 meeting will run from Friday, 29 May, to Tuesday, 2 June, with the plenary session on Sunday, 31 May. The pre-meeting press briefing takes place on Monday, 18 May, and the schedule has been released to the public. In this article, we will examine the congress’s studies most likely to directly affect clinical practice category by category: the 5 showcase studies selected for the plenary, drug-class innovations (new-generation ADCs, bispecifics, oral SERDs, and a pan-RAS inhibitor), targeted therapy plus molecular genetics, ctDNA and MRD biomarkers, the link between GLP-1 and cancer prevention, MCED early-detection tests, and surgery–radiotherapy–lifestyle de-escalation strategies. Let us say it up front: This congress will have one headline: RASolute 302 — the first true RAS inhibitor long awaited in pancreatic cancer reduced the risk of death by 60%, raising median survival from 6,7 months to 13,2 months. A gain of this magnitude is the biggest single-headline shift in pancreatic cancer over the past 15 years.

Study most likely to enter practice rapidly (priority for Turkish oncology practice)
29 May - 2 Jun
Chicago, McCormick Place — Plenary on Sunday, 31 May
— 1. PLENARY SESSION: 5 SHOWCASE STUDIES —

Sunday, 31 May, on McCormick Place’s Main Stage

ASCO’s annual plenary session features the congress’s “showcase” studies. The five selected this year (Late-Breaking Abstracts, LBA) represent all five main axes of modern oncology: perioperative intensification, biologically driven drug repurposing in a rare tumor, targeted adjuvant therapy, next-generation immunotherapy, and direct RAS inhibition.

LBA1 — PROTEUS

Perioperative apalutamide + ADT in high-risk localized prostate cancer

Johnson & Johnson · Erleada (apalutamide) · Phase 3 final analysis

A randomized study testing whether perioperative apalutamide + ADT, before and after radical prostatectomy, is superior to ADT alone in high-risk localized/locally advanced prostate cancer. J&J announced that the results signal “a potential paradigm shift.”

Implications for Turkish practice: For high-risk localized prostate cancer cases discussed in Memorial Göztepe tumor board reports, the question of “the role of hormonal manipulation in curative-intent surgery” could be clarified by the first large randomized dataset. It will directly affect decisions by the Urology + Medical Oncology board.

LBA2 — SARC041

Abemaciclib vs placebo in advanced dedifferentiated liposarcoma

Lilly · Verzenio (abemaciclib) · Based on CDK4 amplification

The interesting aspect of this study is that a CDK4/6 inhibitor known as a breast cancer drug is being tested in a rare sarcoma in which CDK4 amplification is common. In other words, the rationale of matching drugs to tumor biology rather than tumor type.

Implications for Turkish practice: Clinical validation of a biomarker-guided approach to CDK4 amplification in soft-tissue sarcoma tumor board reports; also an important example of tissue-agnostic biomarker matching in rare tumors.

LBA3 — LIBRETTO-432

Adjuvant selpercatinib in resected RET fusion-positive NSCLC

Lilly · Retevmo (selpercatinib) · Stage IB-IIIA · Adjuvant setting

Adjuvant selpercatinib after curative surgery in patients with early-stage NSCLC and a RET fusion. Positive topline results were announced in February 2026; an OS signal is awaited. It will extend the line established by ADAURA (osimertinib, EGFR) and ALINA (alectinib, ALK) to RET.

Implications for Turkish practice: Adding selpercatinib after curative surgery for early-stage lung adenocarcinoma with a RET fusion could become standard. The practical message: the need for comprehensive molecular testing at diagnosis — without waiting for recurrence — will become even stronger. This reinforces the requirement for a comprehensive NGS panel with every new NSCLC diagnosis at Memorial Göztepe.

LBA4 — HARMONi-6

Ivonesimab + chemotherapy vs tislelizumab + chemotherapy in untreated advanced squamous NSCLC

Akeso/Summit · Ivonesimab (PD-1 × VEGF bispecific) · Developed in China

Ivonesimab is a bispecific antibody that targets PD-1 and VEGF simultaneously. PFS data were presented at ESMO 2025; at ASCO, overall survival findings will be announced. This is clinically important because progress in squamous histology has historically lagged behind nonsquamous histology.

Implications for Turkish practice: The possibility that a bispecific antibody + chemotherapy combination could replace the PD-1 monotherapy standard in squamous NSCLC. A candidate for a new frontline standard for Memorial Göztepe lung oncology boards.

LBA5 — RASolute 302 — THE CONGRESS HEADLINE

Daraxonrasib vs chemotherapy in previously treated metastatic PDAC

Revolution Medicines · Daraxonrasib (RAS-ON pan-RAS inhibitor) · 501 patients

Directly hitting RAS, historically considered an “undruggable” target, the first positive Phase 3 data. Daraxonrasib is an oral, multiselective, GTP-bound RAS-ON inhibitor covering KRAS G12 mutations (G12D, G12V, G12R) and wild-type RAS. The study’s principal investigator is Brian M. Wolpin, Dana-Farber Cancer Institute.

📊 Median OS: 13,2 months vs 6,7 months — HR 0,40, p < 0,0001 (60% reduction in the risk of death)

Implications for Turkish practice: In pancreatic tumor board reports at Memorial Göztepe and other oncology centers, it will completely rewrite the 2nd-line treatment scenario. An OS improvement of this magnitude in mPDAC has not been seen for more than a decade. It received FDA Breakthrough Therapy Designation and a Commissioner’s National Priority Voucher — approval is expected in late 2026/early 2027.

Bonus — outside the plenary but in the spotlight: LBA6007 — Ultra-low-dose immunotherapy + oral metronomic chemotherapy vs paclitaxel–carboplatin in platinum-sensitive recurrent/metastatic HNSCC (Phase 3). For health systems with limited resources, there is considerable potential to offer a low-cost alternative — noteworthy for Türkiye’s SGK-based practice.
— 2. DRUG-CLASS INNOVATIONS —

Pan-RAS, New-Generation ADCs, Bispecifics, Oral SERDs

Pan-RAS inhibition

Discussed above with RASolute 302. A shift from targeting a single KRAS G12C variant (sotorasib, adagrasib) to the entire RAS G12 spectrum. An effective RAS inhibitor in PDAC will also accelerate parallel clinical research in lung and colorectal cancers. Similar Phase 3 studies across all non-KRAS G12C variants are expected to open after ASCO.

The ADC wave — new payloads, new targets

Abstract
3505
Trastuzumab rezetecan vs SOC, HER2+ chemotherapy-refractory advanced colorectal cancer (Phase 3). HER2 amplification is now a subgroup that must be actively targeted in CRC. The second major randomized dataset in this field after DESTINY-CRC02.
Abstract
4008
Izalontamab brengitecan vs chemotherapy, recurrent/metastatic esophageal squamous cell carcinoma (Phase 3). A new-generation B7-H3-targeted ADC developed in China.
LBA1003
Izalontamab brengitecan vs physician’s-choice chemotherapy, mTNBC. After sacituzumab govitecan, the second-generation ADC landscape in mTNBC is taking shape.

HARMONi-6
Ivonesimab — PD-1 × VEGF bispecific. Discussed above in the plenary section.
Abstract
7507
MajesTEC-9 — Teclistamab monotherapy vs PVd/Kd in R/R multiple myeloma. According to topline data, teclistamab reduced the risk of death by 40% and the composite risk of progression or death by 71%. It could redefine the second-line standard in MM. Critical for Memorial Göztepe hematology boards.
LBA7506 —
SUCCESSOR-2
Mezigdomide + carfilzomib + dexamethasone vs Kd in R/R MM. A new-generation CELMoD (Cereblon E3 Ligase Modulator Drug); successor to the lenalidomide/pomalidomide class.
LBA7000 —
frontMIND
Tafasitamab + lenalidomide + R-CHOP in newly diagnosed DLBCL. The second anti-CD19 intensification strategy after Pola-R-CHP. The frontline DLBCL paradigm may change again.

Oral SERDs and the hormonal endocrine backbone

LBA1006 —
persevERA BC
Giredestrant + palbociclib vs letrozole + palbociclib in 1L ER+/HER2- advanced breast cancer. Failure to meet the primary endpoint was announced in March 2026. The data presentation will reopen the question of how much clinical difference the endocrine backbone combined with CDK4/6 makes.
LBA1007 —
SERENA-6
Camizestrant for an ESR1 mutation emerging during 1L endocrine therapy; final PFS2 data. A practical prototype of the shift toward “molecular progression = clinical progression”. Details will follow in the ctDNA monitoring section.
Abstract

Other important drug-class studies

LBA5007 —
TALAPRO-3
Talazoparib + enzalutamide vs placebo + enza, HRR-mutant metastatic castration-sensitive prostate cancer (mCSPC). Achievement of the primary endpoint was announced in March 2026. The PARP + AR pathway combination is moving into the frontline in prostate oncology.
LBA2007 —
ESAONA
Asandeutertinib (3rd-generation EGFR-TKI) vs osimertinib, EGFR-mutant NSCLC with brain metastases. Intracranial activity and a head-to-head comparison with osimertinib are rarely available data. At Memorial Göztepe lung oncology boards, this may affect treatment selection for EGFR+ patients with brain metastases.
LBA9503 —
OptimUM-02
Darovasertib + crizotinib vs physician’s choice in HLA-A2-negative metastatic uveal melanoma (Phase 2/3). For the patient group not covered by tebentafusp, after a long wait, the first genuine randomized option.
— 3. GENETICS & TARGETED THERAPY —

Areas in which molecular matching is evolving

ASCO 2026 offers data across all axes of molecularly guided oncology:

TARGETED THERAPY SUMMARY — STUDY × MOLECULAR TARGET MATRIX

  • RET fusion — LIBRETTO-432 (LBA3): It will strengthen the place of targeted therapy in the adjuvant setting. Adding selpercatinib after curative surgery in early-stage lung adenocarcinoma with a RET fusion could become standard.
  • RAS mutations — RASolute 302 (LBA5): A broad spectrum in PDAC, including RAS G12 (particularly G12D, G12V, G12R) and wild-type RAS. Other agents targeting KRAS G12D are also advancing rapidly.
  • BRAF V600E mCRC — Abstract 3504 SWOG S2107: Encorafenib + cetuximab ± nivolumab in MSS BRAF V600E metastatic CRC (Phase 2). After BREAKWATER, this will show the value of adding immunotherapy in this patient group.
  • HRR mutation — TALAPRO-3 (LBA5007): It tests the frontline role of a PARP + ARSI combination in mCSPC patients with BRCA1/2 and other HRR alterations.
  • HER2+ colorectal — Abstract 3505: With trastuzumab rezetecan, HER2 amplification is gaining strength as a subgroup that should be actively targeted in CRC.
  • CDK4 amplification — SARC041 (LBA2): Dedifferentiated liposarcoma commonly carries MDM2/CDK4 amplification; an important example of how “tissue-agnostic biomarker matching” repurposes drugs in rare tumors.
— 4. BIOMARKERS: ctDNA, MRD, MIRNA, LIQUID BIOPSY —

Saturday, 30 May, 08:00 — ctDNA Clinical Science Symposium

The central biomarker stage at this congress will be the session on the morning of 30 May, “Clinical Science Symposium — ctDNA in Clinical Practice: From Detection to Clinical Decision-Making” . NHS-Galleri (LBA100) has been placed in this session. For drozdogan.com, this session is the main reference for the content calendar of our ctDNA monitoring series.

LBA1007 —
SERENA-6
Camizestrant for an ESR1 mutation emerging during 1L endocrine therapy. The first pivotal Phase 3 study proposing an endocrine therapy switch when ctDNA detects an ESR1 mutation — before disease progression. A shift toward “molecular progression = clinical progression.” The second major example of ctDNA-guided clinical decision-making alongside the FDA approval of atezolizumab/Signatera on 15 May 2026.
LBA5003 —

— 5. CANCER PREVENTION: GLP-1, LIFESTYLE, WEIGHT —

One of ASCO’s most prominent thematic blocks this year

Half of the ASCO 2026 press briefing is devoted to lifestyle and metabolic intervention studies. This indicates a dramatic shift in the scientific community’s priorities in cancer prevention. In particular, GLP-1 receptor agonists (semaglutide, tirzepatide) are being systematically examined within a cancer-prevention framework.

GLP-1 receptor agonists — patterns of oncological effects

Abstract
629
Real-world treatment patterns and mortality associated with GLP-1 RAs in breast cancer patients.
Abstract
3143
Propensity-matched analysis of whether GLP-1 RAs mitigate cancer progression in seven solid tumors.

GLP-1 + CANCER — CONCEPTUAL FRAMEWORK

Together, these four studies will form the first major nonrandomized but matched synthesis positioning GLP-1 RAs in oncology for both prevention and metabolic support during active treatment . The practical implication for Memorial Göztepe: a scientific basis for assessing the role of GLP-1 RAs in treatment plans for obesity-related cancers (breast, colorectal, pancreatic, liver, endometrial, and kidney).

Important note: ASCO 2026 data should be read alongside our article on the obesity–cancer link and our article on Akkermansia muciniphila / weight maintenance (May 2026). When these three sources are considered together, “metabolic oncology” becomes visible as a new subdiscipline.

Weight, nutrition, exercise, digital health

— 6. EARLY DETECTION: MCED TESTS AND SCREENING —

NHS-Galleri (LBA100) — The congress topic most likely to attract public attention

NHS-Galleri will probably be the topic attracting the greatest public interest at this congress. A pivotal study that will determine the future of Multi-Cancer Early Detection (MCED) tests.

LBA100 — NHS-GALLERI

Annual MCED screening + standard screening vs standard screening alone in 142.000+ participants

GRAIL/NHS · Participants aged 50-77 · Prospective, over 3 years

Topline data announced in February 2026: a statistically significant reduction in combined stage III-IV cancers — the primary endpoint — was not achieved. However, a favorable trend over time was reported in a subset of 12 cancers (anal, bladder, colorectal, esophageal, head and neck, liver/biliary tract, lung, lymphoma, myeloma, ovarian, pancreatic, and stomach).

Detailed data and subset analyses will be presented at ASCO; they will directly affect the future of population-based MCED policy.

Implications for Turkish practice: A strong topic for an analytical blog post on drozdogan.com; there is a gap in Turkish-language MCED content with a Turkish perspective . Once NHS-Galleri results are detailed, they will provide the essential data for discussing whether MCED tests should enter Turkish clinical practice.

LBA10509 —
PATHFINDER 2
Safety and performance of the Galleri test in a real-use population (a registrational study). Rapid Oral Abstract — Prevention, Risk Reduction, and Genetics session.
GRAIL
Implementation

— IMPACT MAP FOR TURKISH ONCOLOGY PRACTICE —

Among hundreds of abstracts, those that will directly affect Turkish clinical practice and the drozdogan.com content calendar are the following 6 studies:

1
RASolute 302 — Pancreatic tumor board prescriptions will be rewritten
The 2nd-line mPDAC scenario is changing completely. Turkish-language pancreatic oncology content needs reframing.
2
LIBRETTO-432 — Adjuvant standard for resected RET+ NSCLC
It raises NGS testing in early-stage lung adenocarcinoma from a “recommendation” to a “requirement.” A practical message for the NGS panel standard.
3
PROTEUS — Hormonal intensification in high-risk prostate cancer
It clarifies the role of hormonal manipulation in a curative-intent approach. Urology–Medical Oncology board decisions gain depth.
4
SENOMAC — The axillary dissection decision in breast surgery
Clarification after AMAROS+SINODAR-ONE. A direct reference for Memorial Göztepe breast surgery boards.
5
NHS-Galleri — The gap in Turkish-language MCED content
A strong topic for an analytical drozdogan.com blog post. There is a substantial gap in Turkish-language content on the place of MCED tests in Turkish clinical practice.
6
MajesTEC-9 — Second-line MM in hematology board reports
Teclistamab monotherapy vs PVd/Kd: a 40% reduction in the risk of death. It will redefine the second-line MM standard.
⚡ DROZDOGAN ACADEMY — CONCEPTUAL FRAMEWORK
⚡ WHY DOES IT MATTER?

ASCO 2026 (29 May - 2 June, Chicago McCormick Place) is the annual showcase of modern oncology’s progress on multiple fronts. 5 plenary studies (PROTEUS prostate, SARC041 liposarcoma, LIBRETTO-432 NSCLC, HARMONi-6 squamous lung, RASolute 302 pancreas) + 100+ LBAs + GLP-1 cancer prevention as a priority theme + debate on the future of MCED + ctDNA-guided clinical decision-making. The most direct impact on Turkish oncology practice: the RASolute 302 pancreatic revolution, LIBRETTO-432 adjuvant RET+ NSCLC, SERENA-6 ESR1 mutation-guided endocrine switching, SENOMAC breast axillary surgery, NHS-Galleri MCED debate, and MajesTEC-9 teclistamab in MM.

✅ BIGGEST SUCCESSES — EVIDENCE

RASolute 302: An effect unseen in pancreatic oncology for more than 15 years — OS HR 0,40, median OS 13,2 months vs 6,7 months, p<0,0001. MajesTEC-9 teclistamab: In R/R MM vs PVd/Kd, a 40% reduction in the risk of death and a 71% reduction in the risk of progression or death. SERENA-6: ctDNA-guided endocrine switching, a prototype of “molecular progression = clinical progression.” LIBRETTO-432: Adjuvant selpercatinib is EFS-positive in RET+ NSCLC. HARMONi-6 ivonesimab: Bispecific frontline data in squamous NSCLC. NHS-Galleri: In 142.000+ participants, the MCED primary endpoint was not met, but a favorable subset trend will affect the future of population-based MCED policy.

⚠️ LIMITATIONS AND POINTS REQUIRING ATTENTION

The plenary press announcements provide topline data; full data and subgroup analyses will be disclosed in ASCO presentations. The RASolute 302 toxicity profile (especially rash — a typical adverse effect of the RAS-ON inhibitor class) and KRAS G12 subgroup data are not yet complete. NHS-Galleri did not meet its primary endpoint — a “not yet” message for MCED policy. PersevERA BC did not meet its primary endpoint — the oral SERD class is seeking its place. For SGK reimbursement decisions in Türkiye, presenting clinical evidence is sufficient, but budget-impact assessment will also be needed. Access to new drugs through private funding at centers such as Memorial Göztepe will be gradual during 2026-2027.

🩺 IMPLICATIONS FOR CLINICAL PRACTICE

Clinical notes to be added to Memorial Göztepe board reports after ASCO 2026: Pancreatic oncology — Assessing daraxonrasib for 2nd-line mPDAC (FDA approval expected in late 2026/early 2027, access through private funding). NSCLC — Adjuvant selpercatinib for resected RET+ early-stage disease; the requirement for comprehensive NGS panels at each new NSCLC diagnosis grows stronger. Breast oncology — The ESR1 mutation-guided camizestrant option with SERENA-6 (an extension of drozdogan.com’s May 2026 ctDNA article). Clarification of axillary surgery decisions with SENOMAC. Prostate — Hormonal intensification for high-risk localized disease with PROTEUS; PARP+ARSI combination for HRR-mutant mCSPC with TALAPRO-3. Hematology — 2nd-line teclistamab in R/R MM with MajesTEC-9. For international patient coordinators (UHM, Memorial GMÜ): the new standards must be included in reports for high-risk patients from Eastern Europe/Russia, Central Asia, the Caucasus, and the Middle East.

❓ QUESTIONS FOR THE FUTURE

When will similar Phase 3 results for RASolute 302 and other pan-RAS inhibitors (RMC-7977 and others) emerge in lung and colorectal cancers? Will FDA approval of daraxonrasib come in late 2026 or early 2027? How many months later will SGK decide on reimbursement in Türkiye? Will NHS-Galleri and PATHFINDER 2 data affect the entry of MCED tests into population screening programs? Will the Turkish Ministry of Health launch a national pilot program for MCED? When will the first randomized controlled studies of GLP-1 RAs in oncology be designed? Which tumor types beyond breast cancer will the SERENA-6 paradigm (ctDNA-guided proactive treatment switching) extend to? When will a single standard NGS panel for lung adenocarcinoma become a national guideline? How quickly will Memorial Göztepe and other tertiary oncology centers update their content calendars, board report structures, and international patient treatment plans after ASCO 2026?
